HP Ingredients’ LJ 100 ® Receives Safety Confirmation
by INTERTEK And Health Canada NPN License

Tongkat Ali root extract deemed safe at 200 mg per day, receives Health Canada NPN License.

HP Ingredients, Bradenton, FL, a manufacturer and supplier of organic non-GMO and sustainably wild-harvested patented herbal extracts, announced two significant regulatory and safety reviews that will greatly help marketers of its LJ 100 Ingredient for sports performance, maintaining high free testosterone levels, enhancing sexual function and promoting overall well-being, mood state and hormonal health.

Barry Lynch, director, Food and Nutrition Group, Intertek Scientific & Regulatory Consultancy (Intertek), reviewed a total of 56 studies (human clinicals and animal studies encompassing safety, mechanism of action and efficacy in a range of sexual and reproductive support issues and concluded, "The available information supports the safety of LJ 100 at the intake level of 200 mg per day. "

In his written review (dated Feb. 16, 2016), Mr. Lynch asserted, "The components of this extract are consistent with the specifications established in the Malaysian Standard. Emphasis was placed on assessing the safety of incorporation of this specific aqueous extract of tongkat ali root into dietary supplement products and, further, to determine a dose not likely to cause significant adverse effects. Specifically, Intertek evaluated the safety of a proposed dosage of 200 mg/day in healthy adults."

The report also pointed out that researchers have reported increased serum testosterone and DHEA concentrations in several clinical trials in which LJ100 was administered at doses of generally 200 mg/day or more. Other researchers have reported no such effect on testosterone and/or DHEA concentrations.

Mr. Lynch concluded that based on an exhaustive review of the available data that show a lack of serious adverse effects at doses of 200 to 600 mg/day, there is no indication that consumption of 200 mg/day of LJ100 would be associated with adverse effects.

"This review by INTERTEK further supports the safety of LJ 100 brand of standardized aqueous Eurycoma longifolia extract specifically and does not encompass generic Tongkat Ali alternatives,” said Annie Eng, founder and president of HP Ingredients.

In addition, Health Canada has recently concluded that HP Ingredients' application for its proprietary Eurycoma longifolia ingredient known as LJ100 "is in compliance pursuant to section 7 of the Natural Health Products Regulations (NHPR)," and has thus issued product license NPN #80067049 for LJ100 capsules. Further, the following claims are now allowed to be made for Canadian LJ100 products (100 mg and 200 mg):

  • “Eurycoma longifolia helps support healthy sexual interest."
  • "Eurycoma longifolia helps promote testosterone production."
  • "Eurycoma longifolia supports testosterone production."
  • "Eurycoma longifolia is a source of/provides antioxidants.”

"Our LJ 100 is one of the very few testosterone ingredients currently available on the market that has gone through this type of rigorous independent and third party safety review," said Hame Persaud, executive vice president, HP Ingredients. "At a time when free testosterone enhancing products are under intense scrutiny, this should lay to rest any concerns regarding the safety of LJ 100 for oral dosage use in dietary supplements at its clinically evaluated levels."

Bradenton, FL -- Annie Eng, CEO of HP Ingredients Corp., has announced that the company's NeuroActin® was awarded a patent -- US 2015/0352075 A1 -- for its potential use in brain and cognitive support supplements.

The patent, "Treatment of Alzheimer's and Cognitive Impairment with Andrographolides," describes how Andrographolide (ANDRO), a labdane diterpene and a major component of Andrographis paniculata (NeuroActin®) was found to prevent and reverse neuropathology and stimulates neurogenesis.

GSK-3b is an enzyme that regulates the Wnt signaling pathway; it suppresses brain activity by reducing the flow of chemical messages sent between brain cells. The Wnt signaling pathway is essential in the central nervous system as it participates in neural tube formation and mid-brain development. When the Wnt pathway is blocked, this reduces healthy neurogenesis and impairs learning and memory function. Patent co-inventor Nibaldo Inestrosa states, "We found that Andrographolide activates Wnt signaling and inhibits GSK-3b. In doing so, Andrographolide induces post-synaptic proteins and synaptic function, improves memory and spatial learning in APP Alzheimer model mice, and stimulates neurogenesis.”

Andrographolide is a potent anti-inflammatory agent isolated from the plant Andrographis paniculata. It has been reported to inhibit NF-κB at the DNA binding. “In our research, we have found that andrographolide reduces neuro-inflammation, protects against neuron cell loss, reduces amyloid-beta peptide levels and prevents the breakdown of the tau protein," explains patent co-inventor Dr. Juan Hancke.

In a healthy brain, microtubules are the transport system, organized in orderly parallel strands, where food molecules, cell parts and other key materials travel. Tau protein stabilizes microtubules. Beta amyloid causes tau protein to collapse into twisted strands called tangles. When Tau proteins are defective, they no longer stabilize microtubules properly. Nutrients and other essential supplies can no longer move through the cells, which eventually cause neuronal cell death. “Andrographolide prevents tau hyperphosphorylation and significantly reduces the amyloid deposits in the brain.” states Dr. Hancke.

In 2015 alone, Inestrosa and Hancke had published a total of three research papers about Andrographolide and Alzheimers. The latest research titled “Andrographolide Stimulates Neurogenesis in the Adult Hippocampus” was published on Neural Plasticity in December 2015. This presents a significant breakdown as it shows the potential of Andrographolide to induce generation of newborn neurons, increases density of immature neurons, increases percentage of the granular cell layer and more complex morphology.

Eng comments, “Typically, a 30-minute aerobic exercise, such as running, has positive effects on brain structure and function, for example, the generation of neurons (neurogenesis) in the hippocampus, a brain structure important in learning. We are thrilled to learn of the potential of Andrographolide in stimulating neurogenesis. While our specific in vitro experiments entail markers for Alzheimer's disease, our research indicates a much broader potential for Andrographolide to treat other forms of cognitive impairment."

In addition, she says, “Although the dietary supplement industry cannot produce supplements to treat a disease state, our patent supports the concept of protecting the brain's structure, promoting better memory and sharper mind, and hence, its healthy functioning."

NeuroActin® is an Andrographis paniculata extract standardized to contain not less than about 50% (w/w) andrographolide, 14-deoxyandrographolide, neoandrographolide and a combination thereof.

Is This The Powerful New Anti-Inflammatory Ingredient?

In the recent Joint Health Article by Natural Products Insider, inflammation is characterized by "fluid buildup, heat and redness, joint pain, stiffness and loss of joint function."

The typical conventional response is to take a non-steroidal anti-infammatory drug (NSAID), asprin, naproxen, or ibuprofen, or corticosteroids (prednisone), which, if repeated regularly, can cause negative side-effects, such as liver and kidney problems, bleeding in the stomach and intestines, and even high blood pressure. The newer class of COX-2 inhibitors (such as Celebrex), however can also become problematic, increasing cardiovascular events by completely shutting down COX-2, thereby suppressing production of the blood-clotting hormone prostacyclin and other cardio-protective enzymes.

For all its benefits, there is a down-side to getting physically fit. Annie Eng, CEO of HP Ingredients in Florida said as more Americans are getting in shape by regular workouts to fitness centers, ironically, they often lose more than just weight; they often lose joint integrity via the physical stress of repetitive motion.

The Claim:

An ingredient with a history of use in Chinese and ayurvedic medicines, is from Andrographis paniculata, an annual herbaceous plant. The Verdict:

ParActin® administered at 100 mg per day for 14 weeks decreased the intensity of joint pain in patients with rheumatoid arthritis (RA) compared to placebo in a 2009 study.¹ Those who took the botanical also had fewer tender joints and number of swollen joints. Patients treated with ParActin also showed improvement in C-Reactive Protein from above normal to a normal level, and demonstrated an ability to normalize immunoglobin (Ig) A and IgM. The reduction in IgA and IgM is beneficial as there is positive correlation between the grade of cartilage damage.

In another study from 2013, 300 mg per day of ParActin® for four years significantly improved total grade of tender joints and quality of life in patients with various rheumatoid conditions.² In addition, researchers found a significant reduction in Rheumatoid Factor and CRP, pain and fatigue.

New Men's Fitness Ingredient Proven by MIT To Enhance Sports Performance

In the sports and fitness world, testosterone should not be ignored. When testosterone levels are in healthy, youthful ranges it contributes to muscle mass and strength gains by increasing the synthesis of muscle protein.

Testosterone plays a key role in its its effect on male sexual health; it’s essential in producing erections. Testosterone also stimulates metabolism, promotes fat burning, builds muscle, enhances mood, and increases energy in both men and women.
For the increasing number of middle-aged men, a majority of whom produces less of this hormone, the idea of adding a natural testosterone-production ingredient into fitness formulas may be a very attractive concept.

Interestingly, according to Todd Schroeder, Ph.D., a researcher who focuses on exercise and hormones in older men at the University of Southern California, research does show that both endurance and weight-training exercise will temporarily increase testosterone production--anywhere between 15 minutes and an hour after cessation of the fitness routine.

Testosterone production for men begins to slowly decline around age 30. Over time, as this decrease continues, symptoms emerge, including weight gain, decreased muscle mass, mood disorder, and decreased sex drive. As men age the Leydig cells that secrete testosterone begin to wear away. Because of this, between 40 and 70, the average man loses nearly 60 percent of his tesosterone! Other lifestyle factors, such as overtraining, stress and alcohol, can also hasten the deterioration of Leydig cells and cause testosterone levels to drastically decline.

By the time a man turns 60, only about 2 percent of testosterone is free. The rest is bound by the sex hormone binding globulin (SHBG), which renders testosterone inactive by binding it to fat cells. When testosterone is bound, it can’t be used in the body. Free testosterone is responsible for providing the body with energy and enhancing sex drive.

When testosterone production is restored to a more vigorous rate, men will be able to more efficiently lose body fat. Testosterone is key in the regulation of insulin, fat metabolism, and glucose; so it stands to reason that middle-aged men with lower testosterone production begin to accumulate fat. On top of that, adipose tissue converts testosterone into estrogen, which contributes to further decreases in testosterone production.

Currently, several pharmaceuticals directly add testosterone to the body. I believe it is much safer and healthier to provide ingredients such as Eurycoma longifolia—which has been shown to actively encourage the body to produce more of its own—rather than via an exogenous manner.

The researchers found that the supplement group experienced significant increase in fat-free mass, reduced body fat percentages, increased gross muscle power, and significantly increased arm circumference. In a group of men who performed a 24-hour mountain bike event, the LJ100 E. longifolia group showed cortisol levels were 32.3 percent lower than placebo group. Testosterone levels were 16.4 percent higher compared to placebo group. LJ100 promoted an anabolic state while reducing the catabolic state (high cortisol and depleted free testosterone). Results were published in 2006 in The Journal of the International Society of Sports Medicine.

I believe that Eurycoma longifolia such as LJ100 is an effective, safe, and valuable ingredient for fitness formulas, specifically for men over the age of 30. I hope manufacturers feel the same way.

Andrographis Extract Promotes Memory and Cognitive Health?

The traditional Asian herb Andrographis paniculata has attracted attention for applications ranging from joint health to immune health, but new research suggests the herb may also support healthy cognitive function. HP Ingredients (Bradenton, FL) has launched NeuroActin, a patent-pending and proprietary extract of Andrographis paniculata, on the basis of two animal studies into the ingredient’s effects on the brain.

A study1 published in Biochemical Journal explores the effect of andrographolide (ANDRO) on the Wnt signaling pathway of mice and rats through in vivo, in vitro, and in silico analyses. ANDRO is one active component of Andrographis paniculata and the Wnt signaling pathway plays a crucial role in the nervous system. Dysfunction of Wnt signaling “is associated with several neurodegenerative diseases such as schizophrenia and Alzheimer’s disease,” said researchers.

The in vitro kinase assay demonstrated that ANDRO inhibits Glycogen synthase kinase-3 (GSK-3β), an enzyme that regulates the Wnt signaling pathway and neuronal cell function, said researchers. In silico analyses suggest that this inhibition occurs at the substrate-binding site of GSK-3β.

The researchers ultimately concluded that “our results suggest that ANDRO could be used as a potential therapeutic drug for disorders cause by Wnt-signaling dysfunction such as neurodegenerative diseases.”

Another study2 investigated the effect of ANDRO on mice aged 7 and 12 months with AβPP-PS1 mutant transgenes, which are intended to model Alzheimer’s disease (AD) in humans. Researchers found that ANDRO had the effect of reducing harmful amyloid β-peptide (Aβ) levels and tau phosphorylation, both of which contribute to the “underlying neuropathology of AD.”

“ANDRO could play a role in potential preventive therapy during AD progression,” said the researchers.

Researchers in Italy Found Bergamot Lowered Blood Cholesterol
In Some Studies, Daily Supplements Were Effective; Larger Independent Studies Are Needed

The Ache: Common cholesterol-lowering drugs called statins can have unpleasant side effects, including muscle aches and weakness.

The Claim: An extract from a bitter, fragrant citrus fruit called bergamot, commonly known as a flavoring in Earl Grey tea, can lower cholesterol with minimal side effects, some scientists say. It also boosts good cholesterol, reduces fatty deposits in the liver and lowers blood sugar, they add.

The Verdict: Bergamot has been shown to lower cholesterol in at least four human studies published or presented at scientific meetings. But much of the work is authored by a group of Italian researchers who work closely with a company that sells the extract, scientists say. Larger, independent studies in other countries are needed to confirm the results, they say.

Bergamot fruit grows on sunny slopes in Southern Italy and are rich in substances called flavonoids, which likely are the reason for its beneficial effects says Arrigo Cicero, a scientist in the atherosclerosis and metabolic disease research unit of the University of Bologna. But he adds that he feels additional human studies are needed to prove efficacy.

Flavonoids are antioxidants, or substances some scientists believe reduce chemical reactions in the body that damage cells. Earl Grey tea likely contains too little bergamot to have a therapeutic effect, scientists say, though the tea is high in another class of antioxidant called catechins.

Bergamot supplements, often in 500 milligram or 650 milligram capsules or tablets, are intended to be taken before meals. In human studies, a bergamot extract was shown effective at a dose of 500 milligrams to 1,500 milligrams a day, according to published results.

In a month long study of 77 patients published in 2013 in the International Journal of Cardiology, 1,000 milligrams daily of bergamot extract lowered cholesterol from an average of 278 milligrams per deciliter of blood to 191.

The study also found that by adding bergamot researchers were able to reduce by half the dose of the cholesterol drug rosuvastatin without a reduction in efficacy, says study co-author Vincenzo Mollace, professor at the Magna Graecia University in Catanzaro, Italy. Dr. Mollace, a cardiologist, is a paid scientific consultant to Italy’s Herbal & Antioxidant Derivatives Srl, which sells the extract for use in dietary supplements.

“The data looks very good,” says David Frid, a staff physician in the section of preventive cardiology at the Cleveland Clinic.

“But it would be beneficial for this to be reproduced [by another research group] in order to say that in fact this is a true cholesterol-lowering effect.”

“I would tell a patient the caveat is, ‘If you want to try it, you need to be aware that we don’t really know its side effects,’ ” says Dr. Frid. In addition, he adds, statins have been shown to improve health outcomes, such as reducing heart attacks, while bergamot hasn’t yet met that rigorous test.

A glass of bergamot juice tastes like sour grapefruit, making capsules a palatable way to ingest a therapeutic dose, says Annie Eng, chief executive of HP Ingredients Inc., a Bradenton, Fla., company that sells its Bergamonte brand extract to about 30 U.S. dietary-supplement companies.

LJ100 Shown to Have Positive Effects in Male Sexual Performance

Study combined proprietary Eurycoma longifolia with Polygonum minus

BRADENTON, FL -- A new study published in Evidence-Based Complementary and Alternative Medicine demonstrated that administration of a combination of LJ100 (200 mg) Eurycoma longfiolia and Polygonum minus (100 mg) exerted significant sexual performance enhancements over placebo.

The 12-week randomized, double-blind, placebo-controlled, parallel-group study, entitled, "Effects of a Proprietary Freeze-Dried Water Extract of Eurycoma longifolia and Polygonum minus on Sexual Performance and Well-Being in Men: A Randomized, Double-Blind, Placebo-Controlled Study," by Jay K. Udani, et al, was published in January 2014.

A total of 26 healthy men aged 40-65 took part in the study. Subjects who passed screening were randomized. Baseline endpoint data was collected and product was dispensed. Subjects returned for two more visits at week 6 and week 12, where completed diaries and any remaining study product were returned, labs were drawn, and a series of scales and questionnaires were administered. Diaries were completed after each sexual intercourse attempt. Safety labs were repeated at the conclusion of the study at week 12. Further, during the study, subjects were instructed not to alter their current diet and exercise habits.

Those men in the LJ100 group reported a significant increase in sexual satisfaction after 12 weeks, from 15.77 at baseline up to 19.85; the placebo group had a very modest increase, from 12. 36 to 14.29. Erection time from baseline through 12 weeks in the LJ 100 group increased by 12 percent; the placebo group had a moderate increase of almost 5 percent. Further, total testosterone in the LJ100 group increased by 10.36%, compared to only 4.28% in the placebo group.

The authors concluded that the combination of LJ100 and Polygonum minus "significantly improved sexual performance across several clinically important parameters after 12 weeks when compared to placebo. In addition, the product was well tolerated, with excellent safety profiles (identical to placebo) at the administered dose of 300 mg daily for 12 weeks. The results of this study suggest that this product may be of interest to generally healthy middle-aged men who desire improved sexual performance and health.

The study authors explain that erectile dysfunction (ED) occurs as a result of a lack of blood flow to the penis, impairing the ability to achieve or maintain an erection suitable for sexual intercourse. This condition affects approximately 30 million men in the United States and approximately 52% of men aged 40–70 years worldwide. While the incidence of erectile dysfunction increases with age, men at all ages may certainly experience it.

Bergamonte® Shown to Favorably Impact Cholesterol Levels in New Study

Study shows orally administered Bergamonte® extract enhances effects of rosuvastatin

BRADENTON, FL – In a new study published in the International Journal of Cardiology, a team of researchers found that bergamot polyphenolic extract, when taken with the popular cholesterol-lowering statin - rosuvastatin, enhanced the potency of the statin therapy. Previous data showed that Bergamonte extract, rich in brutieridine and melitidine has the ability to inhibit HMG-CoA reductase, which catalyzes the rate-limiting step in mevalonate biosynthesis, a key intermediate in cholesterol metabolism.

"For millions of adults living with elevated cholesterol levels (hyperlipidemia), statins are the pharmaceutical protocol to try to lower those levels into healthy ranges," explains Annie Eng, CEO of HP Ingredients. "However, patients with diabetes and metabolic syndrome do not achieve the recommended cholesterol target with statins alone.”

Based on previous research of bergamot, the researchers investigated the occurrence of an enhanced effect of bergamot extract on rosuvastatin-induced hypolipidemic and vasoprotective response in patients with mixed hyperlipidemia.

The 30-day, open-label, parallel group, placebo-controlled study on 77 patients with elevated serum LDL-C and triglycerides was designed. Patients were randomized to either a placebo (control) group, two groups taking rosuvastatin (10 and 20 mg/daily for 30 days), a group receiving Bergamonte extact alone orally (1000 mg/daily for 30 days;) and a group receiving Bergamonte extract (1000 mg/daily given orally) plus rosuvastatin (10 mg/daily for 30 days).

The researchers found that doses of rosuvastatin and Bergamonte reduced total cholesterol, LDL-C, the LDL-C/ HDL-C ratio and urinary mevalonate in hyperlipidemic patients, compared to control group. 1000mg of Bergamonte and 10mg of rosuvastatin has the same effects in reducing total cholesterol and LDL-C. 1000mg of Bergamonte extract showed much better result compared to 10mg of rovustatin in improving HDL-C and reducing triglycerides.

According to the researchers, the addition of 1000mg of Bergamonte extract to 10mg of rosuvastatin significantly enhanced rosuvastatin-induced effects on serum lipid profile compared to 20mg of rosuvastatin alone. This lipid-lowering effect was associated with significant reductions in trigycerides and increase in HDL-C.

In addition, the cholesterol lowering effect was accompanied by reductions of malondialdehyde, and oxyLDL receptor LOX-1, which correlates to the vasoprotective properties in patients with cardiometabolic risk. The effect of Bergamonte extract in combination with rosuvastatin also resulted in an enhanced expression of PKB levels in PMNs of hyperlipemic patients. These suggest a multi-action enhanced potential of Bergamonte extract in patients on statin therapy.

The authors concluded, "These data are in accordance with previous evidence showing that Bergamonte extract possess antilipidemic and vasoprotective effects both in rat models of hyperlipidemia and vascular disease and in hyperlipidemic patients. In addition, there is a significant additive effect of Bergamonte extract with rosuvastatin, an effect consistent with the further reduction seen in urinary MVA as detected in patients after treatment with both Bergamonte extract and lower doses of rosuvastatin."

These findings reflect those of a previous study, in which Bergamonte extract was found to improve lipid profiles through reduction of LDL, trigyclerides and total cholesterol, increasing HDL-C, and reducing blood glucose in 238 patients in 30 days of daily supplementation.

And Approval from Health Canada

BRADENTON, FL – Annie Eng, CEO of HP Ingredients, has announced that its signature, science-backed ingredient, LJ100 (tongkat ali; Eurycoma longifolia) has obtained not only self-affirmed GRAS status, but an approval from Health Canada.

Under Sec. 201(s) of the FD&C Act [21 U.S.C. 321(s)], a substance is Generally Recognized as Safe (GRAS) if it is generally recognized among experts qualified by training and experience to evaluate its safety either through scientific procedures or common use in food. Self-determination of GRAS status based on a history of safe use in food requires that the substance was commonly used before 1958, and that the safety of the substance is “established by generally available data, including evidence of a substantial history of use of the substance by a considerable number of consumers.

That history of safe use of Eurycoma longifolia dates back approximately 4,000 years ago to a notable series of texts from China.

Authors explain compelling research showing Paractin's powerful role in rheumatoid arthritis relief

The 14-week study gave each patient either a tablet containing 30 mg of andrographolide or a placebo three times a day. According to the authors, the results of the study show a significant reduction at the end of the treatment in the following parameters in the Paractin group: tender joint, number of swollen joints, total grade of swollen joint, number of tender joints, total grade of swollen joints, total grade of tender joints HAQ 0.52 and SF36 (two health questionnaires). The effect was associated to a reduction of rheumatoid factor, IgA, and C4. Patients in another study -- entitled, "Monotherapy with an Andrographis paniculata standardized extract (Paractin®) for the symptomatic relief of different chronic rheumatoid conditions: A prospective case report and long term follow up" -- took two tablets of Paractin® containing 150 mg of standardized A. paniculata extract (90 mg andrographolide per day) for 48 month. The chapter concludes, "Several studies describe a potent anti-inflammatory action of Andrographis paniculata and andrographolide. Andrographolide shows a reduction of the production of pro-inflammatory mediators, such as COX-2, iNOS and cytokines. The molecular mechanism of andrographolide implies the reduction of the activation of transcription factors as NF-κB, AP-1, STAT3 and NFAT and the inhibition of intracellular signaling pathways. A. paniculata standardized extract (30% andrographolide) in clinical trials showed effectiveness for symptom relief and reduce serological parameters in patients with Rheumatoid Arthritis, and the data support a long term treatment similar to other DMARDs."

In one double-blind, randomized, placebo-controlled trial (entitled, "Efficacy of an Andrographis paniculata composition (Paractin®) for the relief of rheumatoid arthritis symptoms: A prospective randomized placebo-controlled trial," the researchers looked at the effects of Paractin® on 60 patients with chronic active rheumatoid arthritis. The 14-week study gave each patient either a tablet containing 30 mg of andrographolide or a placebo three times a day. According to the authors, the results of the study show a significant reduction at the end of the treatment in the following parameters in the Paractin group: tender joint, number of swollen joints, total grade of swollen joint, number of tender joints, total grade of swollen joints, total grade of tender joints HAQ 0.52 and SF36 (two health questionnaires). The effect was associated to a reduction of rheumatoid factor, IgA, and C4. HP Ingredients Issued US Patent For Proprietary Compound, ParActin®

BRADENTON, FL -- Annie Eng, founder and CEO of HP Ingredients, has announced that the company was issued a US Patent for its signature proprietary compound, ParActin®. The patent US #8,084,495 B2, issued on December 27, 2011, is for the unique work: "Composition of Labdane diterpenes extracted from Andrographis paniculata, useful for the treatment of autoimmune diseases, and Alzheimer Disease by activation for PPAR-Gamma receptors."

Peroxisome Proliferator Activated Receptor gamma (PPARy) is the key regulator of the immune and inflammatory response. ParActin® was shown in low doses (25 to 30 mg) to stimulate immune response and at high doses (250 mg) to activate PPARy to promote healthy inflammatory response. When PPARy is activated, then NF-kappaB is inhibited. NF-kappaB, which regulates pro-inflammatory cytokines and pro-inflammatory mediators, is activated in asthma and rheumatoid synovium cells, for example. In vitro and in vivo research showed ParActin® inhibited NF-kB activity and reduced significantly the DNA binding of NF-kappaB.

Autoimmune Mechanism
Autoimmune disorders are expressed in a variety of ways: rheumatoid diseases, psoriasis, systemic dermatomyocytis, multiple sclerosis, lupus erythematosis, and exaggerated immune responses to antigens.

Currently, conventional treatment of autoimmune diseases are the use of immunosuppressant drugs (eg, glucocorticoids, calcineurin inhibitors, anti-proliferatives and anti-metabolites) -- all which may also reduce the immune system's healthy functioning, leaving the patient more vulnerable to infections and cancer. Relatedly, the newer therapeutic advances in immune-suppression (eg, anti-CD3 monoclonal antibodies, anti IL-2 receptor monoclonal antibodies and anti-TNF-alpha monoclonal antibodies) exert associated anaphylaxis reactions and the risk of opportunistic infections such as tuberculosis, as well as other deleterious side effects.

According to the patent inventor, Dr Juan Hancke "The present invention is able to diminish the auto-immune response (which characterizes autoimmune diseases and allergies), alleviating the symptoms and the course of these diseases, while maintaining "immunological tolerance," or a healthy absence of an autoimmune response. We believe that our composition inhibits the synthesis and expression of interferon gamma and interleukin-2, stimulates the PPARy receptor and inhibits the NF kappaB factor. Consequently, we believe these therapeutic compounds in ParActin® selectively reduce over-expression of cytokines, which over-expression is often involved in the pathogenesis of auto-immune diseases."

Alzheimer's Mechanism
Alzheimer’s disease (AD) is characterized by the extra cellular deposition of beta-amyloid fibrils within the brain and the activation of microglial cells associated with the amyloid plaque. The activated microglia subsequently secretes a diverse range of inflammatory products. Earlier research (Kitamura et al) assessed the occurrence of PPARy and cyclooxygenase1, cyclooxygenase-2 in normal and AD brains using specific antibodies and found increased expression of these moieties in brains of Alzheimer's patients. According to Hancke, NSAID s have been shown to be efficacious in reducing the incidence and risk of AD and in delaying disease progression. NSAIDS are PPARy agonists, and research by Combs et al, demonstrated that NSAIDS inhibited the beta-amyloid stimulated secretion of inflammatory products by micoglia and monocytes. PPARy agonists were shown to inhibit the beta-amyloid-stimulated expression of the genes for interleukin-6 and tumor necrosis factor-a and the expression of COX-2.

Until now, concluded Hancke, "there are no antecedents of PPARy agonist compounds isolated from medicinal plants. Whereas, this new composition is able to reduce the pro-inflammatory cytokines production that are increased in autoimmune and neurodegenerative diseases. Additionally, the composition of the present invention has low toxicity, and does not exhibit any harmful side effects."

Two New Human Clinical Trials On Bergamonte and LJ100

Fitoterapia 82 (2011) 309–316 Hypolipemic and hypoglycemic activity of bergamot polyphenols: From animal models to human studies.

Bergamot juice produces hypolipemic activity in rats, though the mechanism remains unclear. Here we investigated the effect of bergamot extract (BPF) in diet-induced hyperlipemia in Wistar rats and in 237 patients suffering from hyperlipemia either associated or not with hyperglycemia. BPF, given orally for 30 days to both rats and patients, reduces total and LDL cholesterol levels (an effect accompanied by elevation of cHDL), triglyceride levels and by a significant decrease in blood glucose. Moreover, BPF inhibited HMG-CoA reductase activity and enhanced reactive vasodilation thus representing an efficient phytotherapeutic approach in combating hyperlipemic and hyperglycaemic disorders.

Andrologia 2011 June 15 Standardized water-soluble extract of Eurycoma longifolia, Tongkat ali, as testosterone booster for managing men with late-onset hypogonadism?

In most countries, millions of people are relying on herbal medicines as remedy for numerous ailments. In South-East Asia, Eurycoma longifolia Jack, also known as 'Malaysian ginseng' or Tongkat ali, is used to combat stress and disease and to improve physical strength. Moreover, the compounds of the roots of this plant are reported to have aphrodisiac and testosterone enhancing effects in the rat. Considering that human studies are not available, 76 of 320 patients suffering from late-onset hypogonadism (LOH) were given 200_mg of a standardised water-soluble extract of Tongkat ali for 1_month. The Ageing Males' Symptoms (AMS) according to the standardised rating scale and the serum testosterone concentration were taken. Results show that treatment of LOH patients with this Tongkat ali extract significantly (P_<_0.0001) improved the AMS score as well as the serum testosterone concentration. While before treatment only 10.5% of the patients did not show any complaint according to the AMS scale and 35.5% had normal testosterone levels, after the completed treatment 71.7% and 90.8% of the patients showed normal values, respectively. Thus, Tongkat ali extract appears to be useful as a supplement in overcoming the symptoms of LOH and for the management of hypogonadism.

New Study Showed Citrus Bergamot Polyphenolic Extract to Lower Total Cholesterol, Lower LDL, Lower Triglycerides, Raise HDL, and Lower Blood Glucose Level

Breakthrough cholesterol lowering citrus from Calabria, Italy

Bradenton, Fl. October 7, 2009 – A new human clinical trial involving 82 dyslipidemic patients showed significant reduction in total cholesterol, LDL, triglycerides, blood glucose level, and significant increase in HDL cholesterol.

Bergamot is the common name of the fruit Citrus bergamia Risso, which grows only in the coastal region of the Ionian Sea in the southern Calabrian region of Italy. Bergamot essential oil is used widely in the cosmetic and perfumery industries. Citrus Bergamot contains high level of naringin, neoeriocitrin, and neohesperidin.

This news comes on the heels of recent published research on Journal of Natural Product, July 2009, which showed bergamot fruit contains rare higher molecular weight flavonoids carrying the 3-hydroxy-3-methylglutaric acid (HMG) moiety. Prof. G. Sindona from Calabria University had identified four flavonoids able to interfere with the natural synthesis of cholesterol in human body, by inhibiting the activity of the first enzyme in the cholesterol synthesis pathway: HMG-CoA reductase (3-hydroxy-3-methyl glutaril-coenzyme A reductase).

The new human clinical trial involved 82 dyslipidemic patients, all with total cholesterol and triglyceride over 250. These patients were given either one or two 500 mg capsule of Citrus Bergamot Polyphenolic Extract for 30 days. At the end of the 30 days, the research showed an average of 31.44% reduction in Total Cholesterol, 42.82% increase in HDL Cholesterol, 38.84% reduction in LDL Cholesterol, 41.54% reduction in Triglycerides, and 21.93% reduction in Blood Glucose level.

Lead researcher, Vincenzo Mollace, PhD, MD, professor at the Faculty of Pharmacy, University “Magna Graecia”, in Catanzaro, Italy was encouraged by the result. “In just four weeks, we saw a significant reduction in total cholesterol, LDL, and triglycerides among these dyslipidemic patients who had elevated cholesterol levels and high risk for CVD. In addition, we are seeing significant increase in HDL, the good cholesterol and reduction in blood glucose as well.” says Mollace.
